Introduction

pUC vectors are a series of plasmid cloning vectors widely used in recombinant DNA technology for gene cloning and related molecular biology work. They are designed to be small, easy to handle, and to carry all essential features of a good cloning vector in a compact form.

Structural Features

  • Size: Each pUC plasmid is about 2,700 base pairs long.
  • Selectable marker (ampR): The ampicillin resistance gene allows only transformed bacterial cells carrying the plasmid to grow on ampicillin-containing medium.
  • Origin of replication (ori): Derived from pBR322, this region enables the plasmid to replicate inside the host cell so that many copies are produced.
  • lac Z gene (reporter region): The lac Z gene from E. coli acts as a reporter; insertion of foreign DNA into its region disrupts the gene, forming the basis of insertional inactivation.

pUC Vector Pairs

pUC vectors occur as pairs, for example, pUC8 & pUC9, pUC18 & pUC19, and pUC118 & pUC119. In each pair, the restriction sites are positioned in reverse orientation with respect to the lacZ promoter.

When the same foreign DNA fragment is inserted into the corresponding restriction site in both members of a pair:

  • one plasmid carries one strand of the inserted gene
  • the other plasmid carries the complementary strand

Because both strands are available in these paired vectors, the inserted gene can be isolated, sequenced and subjected to site‑specific mutagenesis efficiently.

Principle of Cloning (Insertional Inactivation)

The cloning principle in pUC vectors is insertional inactivation of the lac Z gene.

  • Foreign DNA is inserted into the lacZ region or a nearby restriction site.
  • This insertion disrupts the normal function of lacZ, so recombinant clones can be distinguished from non‑recombinant ones using suitable screening methods.

Thus, ampR helps in the selection of transformants, while lacZ is used for screening recombinants.
