- The two strands in DNA are antiparallel i.e. one strand runs in 5’ → 3’ direction whereas the other runs in 3’ → 5’.
- The DNA polymerase synthesizes a new DNA strand in 5’ → 3’ direction only.
- Leading template is synthesized continuously and lagging template is synthesized discontinuously.
- Due to 5’ → 3’ polymerizing activity of DNA polymerase Okazaki fragments are formed only on lagging strand only.
