Solution
Lederberg and Lederberg transferred bacterial colonies from a master agar plate to replica plates using a velvet disc. On plates containing penicillin, only colonies corresponding to resistant colonies on the master plate grew. The matching colony pattern showed that resistant mutants existed before exposure to penicillin; the antibiotic selected them rather than inducing resistance.
